چکیده
directly monitor changes in the twist of nucleic acids Jan Lipfert, Matthew Wiggin, Jacob W.J. Kerssemakers, Francesco Pedaci & Nynke H. Dekker Nature Communications 2:439 doi: 10.1038/ncomms1450 (2011); Published 23 Aug 2011; Updated 27 May 2015 The published version of this article contains two errors. In equation (18), there is a typographical error in the second term; it should read Rcircle 2 Rbead instead of RcircleRbead 2 . The corrected version of equation (18) reads as follows: gy 8pZRbead 1 1=8 ð Þ Rbead=D ð Þ þ 6pZR 2 circleRbead 1 9=16 ð Þ Rbead=D ð Þþ 1=8 ð Þ Rbead=D ð Þ ð18Þ In addition, the y axis of the plot in Fig. 3f was incorrectly scaled. A corrected version of Fig. 3 is shown below. Neither of these errors affects any of the conclusions in the paper, as all calculations were based on the correct power spectrum and the correct version of equation (18). We apologize for any confusion that these errors might have caused.

Freely orbiting magnetic tweezers to directly monitor changes in the twist of nucleic acids
The double-stranded nature of DNA links its replication, transcription and repair to rotational motion and torsional strain. Magnetic tweezers (MT) are a powerful single-molecule technique to apply both forces and torques to individual DNA or RNA molecules. However, conventional MT do not track rotational motion directly and constrain the free rotation of the nucleic acid tether. Here we presen...
متن کاملMagnetic Tweezers for the Measurement of Twist and Torque
Single-molecule techniques make it possible to investigate the behavior of individual biological molecules in solution in real time. These techniques include so-called force spectroscopy approaches such as atomic force microscopy, optical tweezers, flow stretching, and magnetic tweezers. Amongst these approaches, magnetic tweezers have distinguished themselves by their ability to apply torque w...
متن کاملStructural and torsional properties of the RAD51-dsDNA nucleoprotein filament
Human RAD51 is a key protein in the repair of DNA by homologous recombination. Its assembly onto DNA, which induces changes in DNA structure, results in the formation of a nucleoprotein filament that forms the basis of strand exchange. Here, we determine the structural and mechanical properties of RAD51-dsDNA filaments. Our measurements use two recently developed magnetic tweezers assays, freel...
متن کاملNucleosome assembly dynamics involve spontaneous fluctuations in the handedness of tetrasomes.
DNA wrapping around histone octamers generates nucleosomes, the basic compaction unit of eukaryotic chromatin. Nucleosome stability is carefully tuned to maintain DNA accessibility in transcription, replication, and repair. Using freely orbiting magnetic tweezers, which measure the twist and length of single DNA molecules, we monitor the real-time loading of tetramers or complete histone octame...

Living organisms can sense and respond to external and internal stimuli. Response isdemonstrated in many forms including modulation of gene expression profiles, motility, secretion, cell death, etc. Nevertheless, all forms share a basic property: they depend on sensingsmall changes in the concentration of an effector molecule or subtle conformational changes ina protein and invoking the appropri...
